The macrophthalmid crabs, belonging to the subfamily Macrophthalminae of the family Macrophthalmidae, with seven genera and 65 species, are widely-distributed in the middle to low intertidal mudflats and the rocky areas, as well as the subtidal zone, of the Indo-West Pacific. Studies on the macrophthalmids of Taiwan began in 1922, and there are 12 species at present. This study focuses on the taxonomy of Taiwanese macrophthalmids by examination of specimens and review of references. In addition, the DNA barcode is used to confirm species, including the species identification of the mitochodrial 16S rRNA marker. The habitats of each species, as well as the species composition of Taiwan and the adjacent areas, are also studied. Totally 14 species are found in this study, with one new species of Macrophthalmus; three new records (M. crinitus, M. definitus and M. milloti); and two reinstated species (Chaenostoma crassimanus and C. orientale). Molecular evidence shows the minimum interspecific distance is between M. definitus and M. aff. definitus (3.6% in COI and 0.4% in 16S rDNA), which suggest both markers are suitable in species identification in macrophthalmids. The species composition of Taiwanese macrophthalmids is closest to that from the area of Subtropical China (south of Yangtze River). In the types of habitats, some macrophthalmids prefer special habitats, e.g. M. banzai distributes in mudflats around Taiwan; and C. orientale prefers coral reefs with fine sand in northern, eastern and southern Taiwan. In this study, the sequences of mitochondrial DNA (COI and 16S) can help identify closely related species. More markers are suggested to be included to clarify the higher-level relationships.

大眼蟹類屬於大眼蟹科 (Macrophthalmidae) 的大眼蟹亞科 (Macrophthalminae), 共有七屬約 65 種, 廣泛分布於印度-西太平洋中低潮間帶的泥灘與岩礁地區, 甚至包括亞潮帶區域。台灣產大眼蟹類的研究始於 1922 年, 迄今共記錄 12 種。本研究針對台灣產大眼蟹科進行分類探討, 檢視相關的標本形態與文獻紀錄。此外, 並以 DNA 編碼 (DNA barcode) 確認種類, 同時測試粒線體 16S rRNA 標記對大眼蟹類鑑種之適用性。相關種類之棲地類型和台灣及鄰近地區的分布情況亦加以探討。經整理後, 本研究共發現 14 種,其中有新種一種, Macrophthalmus sp. nov.; 新紀錄三種, 包括 M. crinitus、M. definitus、M. milloti; 復活種兩種, 包括 Chaenostoma crassimanus、C. orientale。遺傳距離方面, 最小種間距離在 M. definitus 與 M. aff. definitus 之間 (COI 為 3.6%, 16S rDNA 為 0.4%), 顯示兩種標記對於台灣產大眼蟹種類均具有足夠之解析力。物種組成方面, 台灣與亞熱帶中國 (長江以南) 的大眼蟹組成相最為相似。棲地類型方面, 部分大眼蟹類對棲地有所偏好, 如 M. banzai 幾乎遍布全台之泥灘地; C. orientale 偏好有細沙的珊瑚礁環境, 於北部、台東及墾丁海岸皆有分布。本研究利用粒線體 DNA (COI 和 16S) 的序列分析, 有助於釐清形態相近的大眼蟹物種。未來應使用更多的基因, 以探討屬以上較高階層之親緣關係。
