研究目的
在先前的研究中, 明確的指出辣椒素(反-8-甲基-N-香草基-6-壬烯酰胺)為綠色和紅色辣椒的主要刺激性成分, 對前列腺癌細胞產生深遠的抗增殖作用。 我們希望利用辣椒素(Capsaicin)處理小鼠肺癌細胞(LLC), 探討是否也會有癌細胞生長的抑制作用, 並進一步釐清其作用機轉。
探討方向
(1) 辣椒素對小鼠肺癌細胞株生長之影響。
(2) 辣椒素是否會造成細胞凋亡。
(3) 辣椒素在小鼠肺癌細胞株對粒線體膜電位的影響。
(4) 辣椒素在小鼠肺癌細胞株是否產生活性氧化物。
(5) 辣椒素造成細胞凋亡可能的途徑。
結果
以Trypan Blue計算細胞存活, 不論在48小時或72小時皆顯示隨著辣椒素濃度上升細胞存活有下降趨勢, 而72小時下降趨勢更為明顯。 以流式細胞儀檢測分析,辣椒素濃度200 μM有明顯上升趨勢, 顯示此濃度下會造成細胞凋亡。 以流式細胞儀檢測粒線體膜電位各濃度間無差異, 表示可能並沒有造成粒線體膜電位差的產生,但Bid及Bak蛋白的表現量上確實有明顯變化, 推測可能原因在於辣椒素無法在短時間內造成細胞死亡。 以流式細胞儀檢測活性氧化物質, 並沒有發現有活性氧化物質的產生。 而以西方墨點法分析凋亡蛋白表現量, Bid、Bak、Bcl2、tNOX、Caspase8及PARP有明顯減少, 而p-p53、BIP明顯增加, Caspase12及GADD153無明顯改變, 推論辣椒素引發小鼠肺癌細胞凋亡之途徑可能為經外在途徑刺激活化Caspase 8切割Bid形成tBid而引發內在途徑細胞凋亡。
