Vet Clin North Am Small Anim Pract 40: 221-239, 2010. | 摘要: | 節育被認為是增加肥胖的因子,而肥胖增加了形成糖尿病的風險。雌激素的缺乏可能改變動物食物攝入以及體內代謝從而形成肥胖,增加引起胰島素阻抗。類固醇含量過高也被認為是形成二型糖尿病的一個重要影響因子。類固醇通過改變食物攝入、糖類代謝、脂類代謝而形成胰島素阻抗。三價鉻離子是一種動物體不可或缺的微量元素,其對於糖類、脂質以及蛋白質代謝非常重要。有報導顯示,三價鉻離子可用於減少攝食量,增加代謝率,增加細胞表面胰島素接收器從而提高胰島素敏感性等。因此,本研究欲探討給予三價鉻離子對於節育後或給予外源性類固醇後的血糖及脂質含量的調節作用。本實驗將8周齡的雌性大鼠分為5組(n=6/組),給予生理食鹽水灌胃及生理食鹽水肌肉注射之大鼠為控制組(Control group),進行子宮卵巢摘除手術及生理食鹽水灌胃及生理食鹽水肌肉注射之大鼠為子宮卵巢摘除手術組(OHE group),進行子宮卵巢摘除手術及三價鉻離子灌胃及生理食鹽水肌肉注射之大鼠為子宮卵巢摘除手術加鉻離子組(OHE+Cr3+ group),給予生理食鹽水灌胃及類固醇(Dexamethasone group)肌肉注射之大鼠為類固醇處理組(Dexa-treat group),給予三價鉻離子灌胃及類固醇(Dexamethasone group)肌肉注射之大鼠為類固醇處理加鉻離子組(Dexa-treat+Cr3+ group)。子宮卵巢摘除手術後第二天開始每天給予藥物,之後持續給予12週。每週稱量各組大鼠的體重,分別在8、10、12、14、16、18、20週齡時採集血液測量血糖、胰島素、糖化血色素、膽固醇、三酸甘油酯。在實驗的最後,21週齡是對所有大鼠進行腹腔注射葡萄糖耐受性試驗(IPGTT)。結果顯示,十二週中各組血中葡萄糖含量、糖化血色素含量未出現顯著性差異。類固醇處理組(Dexa-treat group)和類固醇加鉻離子組(Dexa-treat+Cr3+ group)胰島素含量從12週開始顯著高於控制組(Control group),子宮卵巢摘除手術組(OHE group)胰島素含量從14週開始顯著高於控制組(Control group)(P<0.05)。子宮卵巢摘除組(OHE group)從10周開始,膽固醇含量顯著高於控制組(Control group),在第10週時顯著高於子宮卵巢摘除手術加鉻離子組(OHE+Cr3+ group),子宮卵巢摘除手術加鉻離子組(OHE+Cr3+ group)膽固醇含量在第12週時顯著高於控制組(Control group)(P<0.05)。在葡萄糖耐受性試驗中,只有類固醇加鉻離子組(Dexa-treat+Cr3+ group)葡萄糖曲線下面積顯著低於控制組(Control group)和類固醇處理組(Dexa-treat group)。類固醇處理組(Dexa-treat group)胰島素曲線下面積顯著高於控制組(Control group)和類固醇加鉻離子組(Dexa-treat+Cr3+ group),且類固醇加鉻離子組(Dexa-treat+Cr3+ group)高於控制組(Control group)。子宮卵巢摘除手術組(OHE group)胰島素曲線下面積顯著高於控制組(Control group)和子宮卵巢摘除手術加鉻離子組(OHE+Cr3+ group)。綜合以上結果,本實驗顯示給予三價鉻離子能夠緩解子宮卵巢摘除或給予類固醇引起的胰島素阻抗和脂質堆積,增加胰島素對血糖的敏感性。 Neutering was identified as a factor lead to diabetes mellitus. The estrogen deprivation might be the factor to change food intake and metabolism which leads to insulin resistance associated with obesity. Meanwhile, the change of food intake and metabolism may be associated with the secretion of glucocorticoids. Glucocorticoid develops the insulin resistance by exchanging food intake, dyslipidemia and glucose metabolism. Many factors seem like to be involved, such as chromium. Trivalent chromium (Cr3+) is an essential element for carbohydrate, protein and fat metabolism in mammalian nutrition. Many factors seem to be involved, including chromium can reduce food intake, increase metabolism rate, and increase insulin sensitivity by increasing the cell insulin receptors, and so on. The aim of my study will evaluate the influence of trivalent chromium on modulating the level of blood glucose and plasma lipid in the ovariohysterectomized (OHE) rats and the rats treated by dexamethasone. In this study, all 8-week-old SD female rats were divided into 5 groups (n=6): (1) control group, (2) ovariohysterectomy group, (3) ovariohysterectomy with Cr3+ group, (4) Dexamethasone-treated group, (5) Dexamethasone-treated with Cr3+ group. The chromium and dexamethasone were supplied from the day after the OHE performance for 12 weeks. Body weights were recorded weekly. The blood glucose, plasma insulin, HbA1c, cholesterol and triglyceride were measured at 8th, 10th, 12th, 14th, 16th, 18th, 20th week,respectivly. At the end of this study, the intraperitoneal glucose tolerance tests (IPGTTs) were conducted at 21th week. In results, there was no significant difference between the groups in the value of the glucose and HbA1c. The insulin concentrations of Dexa-treat group and Dexa-treat with Cr3+ group were significantly higher than control group from the 12th week, and insulin concentrations of OHE group were significantly higher than control group from 14th week (P<0.05). The cholesterol concentrations of OHE group were significantly higher than that of control group from 10th week and significantly higher than OHE with Cr3+ group at 10th week. The cholesterol concentrations of OHE with Cr3+ group were significantly higher than control group at 12th week (P<0.05). In IPGTTs, the glucose AUC0→120 of Dexa-treat with Cr3+ group significantly lower than those in control group and Dexa-treat group. The insulin AUC0→120 of Dexa-treat group was significantly higher than those in control group and Dexa-treat with Cr3+ group, and this of Dexa-treat with Cr3+ group was significantly higher than that in control group. The insulin AUC0→120 of OHE group was significantly higher than that in control group and OHE with Cr3+ group. According to the results, we concluded that the trivalent chromium supplementation can regulate the insulin effect on blood glucose and lipid accumulation which was induced by ovariohysterectomy or dexamethasone supplement, and slow down the diabetes mellitus development. |
